Genetic diversity and differentiation in Prunus species (Rosaceae) using chloroplast and mitochondrial DNA CAPS markers

Chloroplast (cpDNA) and mitochondrial DNA (mtDNA) were analyzed to establish genetic relationships among Tunisian plum cultivars using the polymerase chain reaction restriction fragment length polymorphism (PCR-RFLP) technique. Two mtDNA regions (nad 1 b/c and nad 4 1/2) and a cpDNA region (trnL-trnF) were amplified and digested using restriction enzymes. Seventy and six polymorphic sites were revealed in cpDNA and mtDNA, respectively. As a consequence, cpDNA appears to be more polymorphic than mtDNA.

Angiopolybia pallens is a swarm-founding wasp that occurs in Atlantic and Amazonian rainforests, which have been isolated from each other by a large arid corridor in central Brazil since the Tertiary. In this study, we used data from polymerase chain reaction-restriction fragment length polymorphism of 16S rDNA to infer some aspects of the genetic diversity and evolutionary history of this social wasp population. Samples collected across the species range in both rainforests showed remarkable differences that separated them into two distinct haplogroups.

Associations between polymorphisms of the GFI1B gene and growth traits of indigenous Chinese goats

This study aimed to investigate polymorphisms of the eighth exon in the GFI1B gene among three indigenous Chinese goat breeds (QianBei Ma goats, GuiZhou white goats, and GuiZhou black goats). Furthermore, association analysis was conducted between these polymorphisms and growth traits. Polymerase chain reaction-single strand conformation polymorphism (PCR-SSCP), direct DNA sequencing, and PCR-restricted fragment length polymorphism (RFLP) were applied to detect polymorphism sites, and a general linear model was used to analyze their association with growth traits.

Polymorphisms of PRLR and FOLR1 genes and association with milk production traits in goats

We investigated the polymorphisms of PRLR and FOLR1 genes in Xinong Saanen, Guanzhong, and Boer goat breeds by DNA sequencing and PCR–RFLP. Two novel SNPs were identified: KC109741: g.62130C>T in the 3ꞌ-UTR of goat gene PRLR, and KC136296: g.7884A>C in exon 3 of goat gene FOLR1. In the three goat breeds, the polymorphism information content was 0.20-0.27 at the g.62130C>T locus. At the g.7884A>C locus, it was 0.36 in Boer goats.

Bovine papillomaviruses (BPVs) are recognized as causal agents of benign and malignant tumors in cattle. Thirteen types of BPVs have already been described and classified into 3 distinct genera. Divergences in the nucleotide sequence of the L1 gene are used to identify new viral types through the employment of PCR assays with degenerated primers. In the present study, a method for identifying BPVs based on PCR-RFLP and DNA sequencing allowed the identification of a new putative Deltapapillomavirus, designated JN/3SP (JQ280500.1).

The sympathetic nervous system plays a major role in blood pressure regulation. Beta 2 (β2) adrenoceptor gene polymorphisms have been associated with hypertension in different populations with conflicting results. We examined the association of three common polymorphisms, Arg16Gly, Gln27Glu, and Thr164Ile, of the β2 adrenoceptor gene in Malaysian hypertensive subjects. A total of 160 hypertensive and control subjects were recruited. Systolic blood pressure (SBP), diastolic blood pressure (DBP), and anthropometric measurements were obtained from each subject.

The aim of this study was to determine the association between a polymorphism in intron 3 of the bovine growth hormone (BGH) gene and growth traits related to the start of the reproductive life of Holstein heifers. This research was conducted using 480 Holstein heifers belonging to eight herds in three municipalities in the Department of Antioquia (Colombia).

The insulin-like growth factor I (IGF-1) gene plays important roles in the growth and body composition of animals. Serum IGF1 concentration has been associated with growth traits in many livestock species. We found a polymorphism of cattle IGF1-TasI locus and analyzed the distribution of alleles in three cattle breeds, including Qinchuan, Nanyang, and Chinese Holstein. PCR-RFLP analysis showed that allele A was the dominant allele. The frequencies of allele A varied from 0.84 to 0.97.

Activation of the peroxisome proliferator-activated receptor g (PPARg) improves insulin sensitivity and inhibits atherosclerosis. Whether PPARg2 Pro12Ala polymorphism affects myocardial infarction is not clearly understood. We investigated a possible association of PPARg2 Pro12Ala polymorphism with obesity and myocardial infarction in Han Chinese in Hohhot, Inner Mongolia, China. We included 121 subjects with myocardial infarction and 137 healthy controls in our study.

Centris spp are oil-collecting solitary bees that are valuable pollinators of crops such as Brazil nut, cashew, and acerola. We investigated the genetic variability of populations of C. aenea in the northeastern region of Brazil. Total DNA was extracted from 59 individuals from 6 locations in the States of Pernambuco and Bahia and a 600-650-bp fragment of the mitochondrial COI/COII region amplified by PCR, followed by digestion with the restriction enzymes DraI and SspI. PCR-RFLP analysis revealed eight different haplotypes among the populations.
