Abstract

Introduction: Pesticides are often associated with various toxic effects in humans, being additionally accountable for a large number of rural suicides or accidental fatalities. In recent times, for better pest management strategies multiclass pesticides with combination of two or more compounds are extensively used. Simultaneous determination of these pesticides in toxicological analyses is challenging and existing approaches rely on chromatographic techniques. Although derivative UV-visible spectroscopy has been used extensively for multicomponent analysis due to its simplicity and cost-effectiveness, however, simultaneous determination of pesticides using the technique remains less explored. Therefore, the present study aims to develop a method for the simultaneous determination of chlorpyriphos and cypermethrin by first order derivative UV-visible spectroscopy. Methods: Human blood samples were taken and spiked with standards of chlorpyriphos and cypermethrin in the ratio 10:1. Then the pesticides were extracted using modified citrate buffer QuEChERS-based method. Microvolume of extracts were analyzed using first order derivative UV-Visible spectroscopy and then processed by the zero-crossing point method. Results: The developed method showed good linearity for both pesticides with correlation coefficients ≥0.99. The method was validated according to ICH Q2(R2) guidelines and the precision study indicated that % RSD was within acceptable limit (<1). LOD for CHL and CYP were 61.905 mg/L and 3.799 mg/L respectively. Whereas, LOQ were 187.593 mg/L and 11.512 mg/L respectively. Conclusion: The present study showed that 1st order derivative UV-visible spectroscopy is a useful analytical technique for routine toxicological analysis of multiclass pesticides. The developed method of analysis offers significant advantages in terms of simplicity, cost-effectiveness, and in agreement of green chemistry principles.
