Abstract

Surface biofunctionalization of titanium implants is essential to improve early osseointegration. Recent advances highlight the role of exosomes in modulating cellular behaviour and enhancing tissue regeneration. The aim of the study is to investigate the influence of exosome- enriched Plasma fraction on surface properties and osteogenic response of titanium discs. Titanium discs were allocated into three groups: untreated control, Plasma fraction treated, and exosome enriched plasma fraction treated. Surface characterization was performed using SEM analysis. Osteoblast response was assessed through RT-PCR expression of RUNX2, Alkaline Phosphatase (ALP), Type 1 Collagen (COL1A1), osteocalcin (OCN), osteopontin (OPN). Surface analysis confirmed successful biofunctionalization without alteration of titanium crystalline structure. Treated groups exhibited increased hydrophilicity and surface energy (p < 0.05). Group 3 demonstrated upregulated expression of osteogenic markers RUNX2, ALP, COL1A1, OPN, and OCN compared to other groups.
