Hyalomma asiaticum tick, an important ectozoic parasite causes tickle, pain, anemia, weight loss, and paralysis in its hosts, which include humans, cattle, sheep, horses, camels, and hares. The 4D8 gene can be a potential vaccine candidate antigen for H. asiaticum. In the present study, we cloned and expressed the 4D8 gene of H. asiaticum from Xinjiang Province. Primers were designed according to the H. asiaticum tick 4D8 gene sequence available in GenBank.

Cloning and sequence analysis of the LOC339524 gene in Sprague-Dawley rats

We cloned the LOC339524 gene in Sprague-Dawley (SD) rats and analyzed the structure and function of the protein encoded by it. Based on the known human LOC339524 gene sequences, the full-length coding sequence of the LOC339524 gene in SD rats was cloned and amplified by the polymerase chain reaction using the complementary DNA of SD rats as a template. Bioinformatics analysis showed that the length of the cloned LOC339524 gene (GenBank accession No. KM224520) was 831 bp and it encoded a deduced protein of 276 amino acids.

The shikimate pathway enzyme 5-enopyruvylshikimate-3-phosphate synthase (EPSPS) is the target of the broad spectrum herbicide glyphosate. A novel aroA gene encoding an EPSPS from Pantoea sp was identified and subcloned into the pET-28a vector to construct the recombinant pET-AroAPantoea sp plasmid. Amino acid sequence analysis indicated that AroAPantoea sp is a class I AroA enzyme. When expressed in Escherichia coli, it conveyed high tolerance to glyphosate.
